Study of determination of activating solution for sperm in wild carp (Cyprinus carpio)
Abstract
In this study effects of activating solution on motility sperm (duration of motility and percentage of motility) of wild carp were investigated. Therefore, after determine optimum pH, effect of those with everyone of Na+, K+, Ca٢+ and Mg٢+ ions were detected, and two saline solution that maximum total period of motility and maximum percentage of motile sperm were selected as activating solution. The activating solution (١) was: (٥٠mM of Nacl, ٣٠mM of kcl, ٣٠mM of Tris, pH = ١٠٫٣١) and activating solution (٢) was: (٥٠mM of Nacl, ٤٠mM of kcl, ٣٠mM of Tris, pH = ١٠٫٦٨). Then effect of these saline solution with Billard's saline solution: (٤٥mM of Nacl, ٥mM of kcl, ٢٫٥mM of Tris,١٩mM of glycine pH= ٨), and Poupard,s saline solution: (٤٥mM of Nacl, ٥mM of kcl, ٣٠mM of Tris, pH=
٨) and distill weter (٠٫٤mM of Nacl) and hatchery water on motility sperm (duration of motility and percentage of motility) of wild carp were investigated. Results of this study showed that maximum total period of motility and maximum percentage of motile sperm were seen in selected saline solution(٥٠mM of Nacl, ٣٠mM of kcl, ٣٠mM of Tris, pH= ١٠٫٣١), (P>٠٫٠٥).
